Kisspeptin is a naturally occurring neuropeptide derived from the KISS1 gene, studied extensively for its central role in regulating the hypothalamic–pituitary–gonadal (HPG) axis. It functions by binding to the GPR54 (KISS1R) receptor, a G protein–coupled receptor expressed in the hypothalamus, where it acts as a key upstream regulator of gonadotropin-releasing hormone (GnRH) secretion. Due to this role, Kisspeptin has become a significant focus of research related to reproductive hormone signaling and endocrine regulation.
More specifically, Kisspeptin is considered a master regulator of reproductive hormone release, as GnRH neurons rely on Kisspeptin signaling to initiate the downstream secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the pituitary gland. Research suggests that Kisspeptin signaling is sensitive to metabolic status, stress, and circadian rhythms, linking energy balance and environmental cues to reproductive function. As a result, Kisspeptin is widely studied in research settings involving puberty onset, fertility mechanisms, and hormonal feedback loops within the endocrine system.
