Kisspeptin is a naturally occurring neuropeptide derived from the KISS1 gene, studied extensively for its central role in regulating the hypothalamic–pituitary–gonadal (HPG) axis. It functions by binding to the GPR54 (KISS1R) receptor, a G protein–coupled receptor expressed in the hypothalamus, where it acts as a key upstream regulator of gonadotropin-releasing hormone (GnRH) secretion. Due to this role, Kisspeptin has become a significant focus of research related to reproductive hormone signaling and endocrine regulation.

More specifically, Kisspeptin is considered a master regulator of reproductive hormone release, as GnRH neurons rely on Kisspeptin signaling to initiate the downstream secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the pituitary gland. Research suggests that Kisspeptin signaling is sensitive to metabolic status, stress, and circadian rhythms, linking energy balance and environmental cues to reproductive function.
